【摘要】Objective To investigate the change of Osterix(Osx) expression in periodontal tissues during orthodontic bone remodeling in rats.Methods 55 six-week-old male SD rats were used as experimental animals.A titanium-nickel closed-coil springwith 40 g force was applied between rat maxillary first molar(M1) and the upper incisors with ligature wires to mesially move M1.0 h,1 h,2h,4 h,8 h,12 h,1 d,3 d,5 d,7 d and 14 d conditions after coiled spring activation were recorded.Upon completion of experiments,the rats were sacrificed for maxillae.Then the specimens were fixed,decalcified,dehydrated,cleared and paraffin-embedded.4 μm serial sections were cut parasagittally.Immunohistochemical staining was carried out with anti-Osx goat polyclonal antibody to examine the expression of Osx.Results In untreated control teeth,Osx was expressed at a low level in the normal periodontal ligament,especially near the cementum.Application of orthodontic force instantly induced a significant increase of Osx expression,which reached its maximumat day 5.Strong positive Osx expression was observed in the periodontal ligament adjacent to root surface at the compression site as well as in those adjacent to root and alveolar bone surface attension site.Osx immunoreactivity was stronger in periodontal ligament at the tension side compared to that at the compression side (P0.01).Besides,the new bone at the tension side also showed an obvious Osx expression.Conclusions The expression of Osx in periodontal ligament was consistent with the change of bone remodeling of periodontal tissue.Osx may play an important role in the periodontal tissue remodeling during orthodontic tooth movement in rats.%目的 观察实验性牙齿移动过程中,牙周膜内成骨转录因子Osterix(Osx)的表达变化,初步探讨Osx与牙齿移动过程中牙周组织骨改建的关系.方法 选用6周龄雄性Wistar大鼠55,随机分为空白对照组、实验加力124812 h135714 d.采用拉簧加力法于上颌右侧第一磨牙建立实验性牙齿移动动物模型,制备牙周组织切片.采用免疫组化及图像分析方法半定量分析Osx在牙周膜的表达变化.结果 在正常对照组,Osx呈弱阳性均匀表达.加力后,Osx着色强度和分布发生一定改变.时间上,加力4h后张力区和压力区牙周膜Osx达即明显增强(P0.01),并逐渐增高至加力5d,双侧Osx表达水平达到最高,后逐渐降低.分布上,在张力区靠近牙骨质和牙槽骨表面及压力区靠近牙骨质表面的牙周膜逐渐呈现强阳性表达,而在发生明显骨吸收的牙槽骨侧无明显着色;自加力3d,张力区阳性强度明显高于压力区.结论 机械力作用下,Osx表达变化具有一定时空规律性,与牙周组织骨吸收和骨形成过程相一致.Osx可能参与了体内正畸牙周组织的骨改建过程,发挥其成骨调控作用.
